Optimization of in vitro seed germination, protocorm growth and seedling proliferation of Vanda Tessellata (Roxb.) Hook. Ex G. Don.

The cultural requirements for in vitro seed germination, protocorm growth and seedling proliferation of Vanda tessellata have been studied. Seed germination media consisted of modified nutrient solutions of Knudson's C, Vacin & Went and half strength Murashige & Skoog, enriched with combinations of organic supplements. For initiation of germination no particular medium preference was observed and in most treatments it ranged from 66 to 73%. However, addition of 0.5% peptone proved inhibitory. The protocorms exhibited differential growth responses in the three culture media and the effects of organic supplements on growth depended on the medium used. In the basal media of Knudson's C and Vacin & Went more than 95 % of the protocorms turned necrotic and the surviving protocorms showed much stunted growth. In contrast, half strength Murashige & Skoog's medium proved effective in decreasing protocorm necrosis and for the improved growth as well. Among the organic supplements, peptone at 0.2% induced maximal growth promotion in the Knudson's C and Vacin & Went media. In addition, peptone decreased protocorm necrosis considerably in all the three media. For further proliferation, the seedlings were cultured on a modified Knudson's C medium containing different concentrations and combinations of BA and NAA.
