DEVELOPMENT AND EVALUATION OF AZITHROMYCIN FLOATING DOSAGE FORM FOR HELICOBACTER PYLORI

Résumé
The objective of the present work was to develop a floating dosage form of Azithromycin tablets intended to extend the gastric residence time (GRT) after oral administration at a particular site and controlling the drug release for achieving controlled plasma level as well as improving bioavailability with localized effect in the treatment of Helicobacter pylori. Tablets containing hydroxy propyl methyl cellulose (HPMC), drug (Azithromycin) and different additives were compressed using wet granulation. The study showed that the tablet composition and mechanical strength have great influence on the floating properties and drug release. The drug release was sufficiently sustained for 6 hours and anomalous diffusion was confirmed. Optimization of the evaluation parameters like bulk density, tapped density, angle of repose, hardness, friability, floating time of tablets and dissolution profile were evaluated for final optimized formulation. The optimized formulation was obtained using Azithromycin (501.60mg), HPMC K-25 (40mg), guar gum (200mg), magnesium stearate Grade I (4mg), magnesium stearate Grade II (8mg), aerosil (4mg), which gave floating lag time less than 30 seconds with a total floating time greater than 6 hours and invitro release profile very near to the intent invitro release profile.
